Mutagenic Research on Folpet

Abstract: 【ABSTRACT】 BACKGROUD & AIM: To assess the mutagenic effects of folpet at chromosomal level. MATERIALS AND METHODS: Comet assay, Ames test, micronucleus test and spermatogonial chromosomal aberration test were performed to analyze the damage of different concentrations of folpet to DNA. RESULTS: Comet test revealed that there was dose-effect relationship on rats peripheral mononuclear cells (PMNC) in vivo and there was significant difference between experiment group and negative control group on human peripheral mononuclear cells in vitro. In Ames test, there was a concentration-related increase over the range tested and a two-fold greater increase in the number of revertant colonies in four strains with or without metabolic activation. No mutagenic effect was seen in micronucleus test and spermatogonial chromosomal aberration test. CONCLUSION: Base pair substitution mutation and frame shift mutation were caused by folpet in Ames test. And DNA breakage in human PMNC might be induced by folpet.
